It is pretty simple to do but you may need to recalibrate the new cell. It will also re calibrate itself after a couple of hours. I also cover when to replace the cell and will show you how to recalibrate the unit. Basically if the salt cell is over five years old or near that age you will start to notice some common problems. The most obvious is that the inspect cell light will stay on solid even after the cell is clean. You will also get a low salt level reading even when the salt level is fine. It is a good idea to get a reading at your local store or with a salt test strip if you think the unit is failing. You may also have problems with low voltage and low amps when the salt cell is generating. This unit had a failed temp sensor inside and needed to be replaced as that is not a repairable part. It is always best to go with the largest salt cell -- the T-15 is rated for 40,000 gallons even if your pool is much smaller. The cell will not have to work as hard and will last longer. Water temperature and the amount of time the system is running also effect cell life. If you don't clean your cell regularly you will also shorten its life. The power unit can also be bad but I haven't noticed any problems with the power unit itself on any of the five Aqua Rite units I service and a couple are over 10 years old or older. They seem pretty durable and trouble free. But just in case you suspect your cell is good and the power unit is bad --try to get a loaner cell from your local pool store or take your cell in to get tested. This is basically a very simple to do and you should encounter no real problems. If you need to talk to tech support call 866-429-7665 and you want the Goldline Division of Hayward. You will see a boost in speed when you find it. I have found that if the antenna is 180 degrees off from the tower I get better upload speeds but bad download. Spin the antenna around and Iv got it. He focuses on the role of the Arp2/3 protein complex in nucleating the growth of the branched actin filaments found at the leading edge of a motile cell. Structural and biochemical studies allowed Pollard and his colleagues to formulate a model for how actin polymerization can provide the force to push the cell membrane forward. In his second talk, Pollard explains why the fission yeast S. pombe is an excellent model to study actin-mediated processes. Fluorescently labeled actin-binding proteins allowed Pollard and his colleagues to follow actual cellular events and compare them with their mathematical model. Adjusting the model so it matched the experimental data provided insight into how actin assembly and turnover could produce the force needed for cells to move or undergo endocytosis. Cytokinesis is the process by which a contractile ring forms around the middle of a cell and squeezes to divide it into two daughter cells. Although it has been known since the 1970s that the contractile ring contains actin and myosin, it turns out that there are over 100 other proteins involved in cytokinesis. In fission yeast, conditional and deletion mutants of many cytokinesis genes are available, many cytokinesis proteins have been labeled with fluorescent tags so they can be followed over time in live cells, and biochemical characterization of proteins has informed computer models. In his third talk, Pollard discusses the data that have given rise to the latest model for how the contractile ring forms and is regulated. Speaker Biography: Tom Pollard is the Sterling Professor of Molecular, Cellular and Developmental Biology at Yale University. He received his BA from Pomona College and his MD from Harvard Medical School. He was a Professor at the Johns Hopkins University School of Medicine from 1977-1996 and President of the Salk Institute in San Diego from 1996-2000. He joined Yale University in 2001. Pollard has been an integral member of the cell biology and biophysics communities for many years. He has been President of both the Biophysical Society and the American Society for Cell Biology, Director of the Physiology course at the Marine Biological Laboratory, and a member of the ASCB Public Policy Committee for 25 years. He has organized numerous meetings, has been a member of many committees and scientific advisory boards, and has been on the editorial boards of at least 10 journals. Together with William Earnshaw, he is the author of the popular textbook Cell Biology. Pollard is a member of the National Academy of Sciences and the Institute of Medicine and a fellow of the American Association for the Advancement of Science. He has received numerous awards including the E.B. Wilson Award from the ASCB in 2005 and the Gairdner International Award in 2006.

Another 1-800-CALLATT commercial. Oft spokesman David Arquette dons a blue chalk mustache and shows how suave he can be by pitching the value of calling collect with 1-800-CALLATT to a woman in a phone booth in a pool hall. Arquette made a lot of 1-800-CALLATT commercials and got a lot of airtime. Most of them were more frenetic than this, this was likely made as a counterpoint to the style that people had become used to. Long distance was big business in the 80s and 90s, with the battle lines drawn first by trying to get people to switch carriers, then by offering "dial around" (10-10) services for cheap calls and finally by offering 1-800 access and other 10-10 services targeting specific portions of the market. 1-800-COLLECT hit gold and then collect calling became a major focus of advertising, even though collect calling had been a rarity and near curiosity for most customers. The long distance industry had been a huge source of ad revenue for TV stations right up there with beer companies but shortly after this, all the angles had been played out and long distance became a low margin industry with little money or reason to advertise. Big long distance finally succumbed to nationwide free long distance (and free roaming) plans on cell phones starting with Sprint PCS' service in1995. Far from a big business, long distance is now a cheap or free service thrown in as a kicker to all-in-one voice plans. This 30 second spot aired in 1999 during a showing of the Monaco GP.

How Does Ting Work? In this video, we'll review the Ting network and discuss their plans. Let's start off by visiting the Ting website. Notice that it says, "The average Ting bill is just $23 per phone, per month". That sounds like a great deal! We're going to check their facts to see if this is actually true. If we click the, "What Would You Save?" button, it takes us to a pricing tool that lets you "see what your bill could be." Keep in mind, the options you select on this screen have nothing to do with your actual bill each month. This is a sales tool. You will never know up front how much Ting will charge. Notice the warning, "Every month is different." This tool lets you select how many lines, or phone numbers, you want to have on your shared monthly pool of minutes, text messages and mobile data. In the first row, the $6 option is preselected for us. This is because Ting charges $6 per line every month. This is not based on your usage at all - you simply pay $6 for each live phone number. That's definitely not the highest line access fee we've seen, but it always seems odd when companies charge you for the privilege to pay them every month. In the second row, there's a note, "The average Ting account uses about 200 minutes per device a month." Since they've provided these numbers, we'll be using their averages for the rest of this example. There isn't a 200-minute option so we'll have to go up to the 500-minute level for an extra $9 per month, bringing your monthly bill to $15. In the third row, it says "The average ting account uses about 300 texts per device a month." Again, there's no option in between 100 and 1000.If you go over 100 text messages, you will automatically be billed for 1,000 text messages at a price of $5. This raises your Ting bill to $20 per month. Lastly, we'll choose the data. "The average Ting account uses about 650MB (or 0.65GB) per device a month." According to their figures, the average Ting customer would go over the 500MB plan by 150MB. This costs you an extra $16 per month, bringing the total bill to $36. That's 156% higher than the $23 listed on their homepage. Now that we've seen the cost for an average customer, let's look at what happens if you run into an emergency. You might end up using more minutes, text messages or data than expected. If we click, "More Minutes" there's a warning that says, "When you use above 2100 minutes you pay $35 plus 1.9 cents for each additional minute." When we click the, "More Text Messages" box, we see a message that reads, "When you send or receive above 4800 text messages you pay $11 plus 1/4 cents for each additional text." Let's look at what happens when you use more data. Here it says, "When you use above 2GB of data you pay $20 plus $10 for each additional gigabyte." These plans could easily get very expensive if someone checked out some videos or downloaded a game. We're not sure how they're able to say, "The average Ting bill is just $23 per phone, per month" when their calculator says the average Ting user uses 200 minutes, 300 text messages and 650 megabytes of data. Using their figures, the average Ting bill looks like it would actually be $36.00 per month. When searching for an answer, we noticed that there are a lot of complaints online. While their plans appear similar to other prepaid companies, they are a post-paid service. Your bill may never be the same from month-to-month so budgeting for your cell phone plan would be difficult. Post-paid billing also allows Ting to run credit checks and submit customers to collections for non-payment. "As you have ignored previous attempts to resolve the outstanding balance on your account we have been forced to suspend your service. Ting came up with a great way to get positive reviews, by actually paying people to give them a good review with a referral link. Apple Watch series 3 unboxing and first impressions review The Apple Watch series 3 has been launched by Apple along with Watch0S 4. The video will focus on the unboxing and I will give you early impressions of the Apple Watch 3 and give you my advice between the Apple Watch series 3 and series 2. This series will focus on the Apple Watch fitness features Heart rate monitoring has got more advanced Apple has removed the heart rate animation from the heart rate app and in its place a graph that gives you more information. There’s also a full heartbeat complication that’ll display your current heartbeat on your watch face. Workout app The most obvious change in the Workout app is the new design, which uses the new vertical scrolling design. But the biggest thing in the Apple Watch workout app update is the addition of triathlon mode. Apple has been since its launch promoting this as a serious fitness watch, but it has missed many features like this and the heart rate changes as already mentioned. I have not tested the new tri-mode out yet, but a quick look it does show some promise. The swimming workout now has an auto lap mode. which your watch will alert you when you have got to the end of a length. The workout app also gets High-Intensity Interval Training supported by the new motion and heart rate algorithms.

HIV infects T-cells, ‘high-jacking’ their cellular machinery and turning them into virus-producing ‘factories’. A small proportion of these cells can create a persisting pool of latently infected cells, which constitute the HIV latent reservoir. There, the virus remains dormant, silent and hidden from patients’ immune defences and from otherwise effective anti-retroviral treatments. In her TEDxUCD 2015 talk Virginie provides an insight into the global research towards eradicating the latent HIV reservoirs, by forcing the virus to ‘wake-up’ and then target and eliminate the viral reservoirs. Dr Virginie Gautier is a Principal Investigator at the UCD Centre for Research in Infectious Diseases in the UCD School of Medicine. Dr Virginie Gautier is a Principal Investigator at the UCD Centre for Research in Infectious Diseases (CRID) in the UCD School of Medicine. Her research programmes focus on the HIV-Host interface and HIV-CURE research. The core expertise of Dr Gautier’s research group lies in molecular and cellular virology, epigenetics, functional genomics and dynamic microscopy. These exploratory approaches have led to the identification of key druggable targets, which are currently under investigation as part of her HIV-CURE Programme which is funded by the Health Research Board, the Irish Research Council and Horizon 2020. Dr Gautier is currently the European co-ordinator of the Horizon 2020 research programme entitled Accelerating HIV CURE research in Europe. She graduated with a BSc (Biochemistry) from the University of Tours (France) and a PhD in Medical Microbiology from University College Dublin. Adoptive T-cell transfer has the potential to overcome the significant limitations associated with vaccine-based strategies in patients who are often immune compromised. Dr. June will discuss how the emerging discipline of synthetic biology-which combines elements of genetic engineering and molecular biology to create new biological structures with enhanced functionalities-can be applied to cancer. He will also talk about the outlook for chimeric antigen receptor (CAR) and T-cell receptor (TCR) T-cell therapies, including managing toxicities and expanding the availability of personalized cell therapy as a promising approach to all hematologic malignancies and solid tumors. Many questions remain in the field of CAR T cells, but the encouraging response rates pave a wide road for future investigation. Carl June is the Richard W. Vague Professor in Immunotherapy in the Department of Pathology and Laboratory Medicine. He is currently Director of the Center for Cellular Immunotherapies at the Perelman School of Medicine, and Director of the Parker Institute for Cancer Immunotherapy at the University of Pennsylvania. He is a graduate of the Naval Academy in Annapolis, and Baylor College of Medicine in Houston, 1979. He had graduate training in Immunology and malaria with Dr. Paul-Henri Lambert at the World Health Organization, Geneva, Switzerland from 1978-79, and post-doctoral training in transplantation biology with E. Donnell Thomas and John Hansen at the Fred Hutchinson Cancer Research Center in Seattle from 1983 - 1986. He is board certified in Internal Medicine and Medical Oncology. He maintains a research laboratory that studies various mechanisms of lymphocyte activation that relate to immune tolerance and adoptive immunotherapy for cancer and chronic infection. In 2011, his research team published findings detailing a new therapy in which patients with refractory and relapsed chronic lymphocytic leukemia were treated with genetically engineered versions of their own T cells. The treatment has also now also been used with promising results to treat children with refractory acute lymphoblastic leukemia. He has published more than 350 manuscripts and is the recipient of numerous prizes and honors, including election to the Institute of Medicine in 2012 and the American Academy of Arts and Sciences in 2014, the William B Coley award, the Richard V Smalley Memorial Award from the Society for Immunotherapy of Cancer, the AACR-CRI Lloyd J. Old Award in Cancer Immunology, the Philadelphia Award in 2012, the Taubman Prize for Excellence in Translational Medical Science in 2014 (shared with S. Grupp, B. Levine, D. Porter), the Paul Ehrlich and Ludwig Darmstaedter Prize (shared w J. Allison), the Novartis Prize in Immunology (shared with Z. Eshaar and S. Two teams at Harvard Medical School reported in separate papers in the May 21, 2015, issue of Cell that their labs have developed high-throughput techniques to quickly, easily and inexpensively give every cell in a sample a unique genetic barcode. This allows scientists to analyze complex tissues by genetically profiling hundreds of thousands of individual cells instead of having to rely on an average of the entire sample. Previously, she was VP and Scientific Director at the New York Academy of Sciences (NYAS) and, before that, held editorial roles at the Journal of Clinical Investigation and Nature Medicine. Stacie Grossman Bloom: So, you know, in this country, I think there’s a big emphasis now on trying to improve STEM: science, technology, engineering, and mathematics education in younger people. I think that in order to pursue a career in research, there have to be role models and incentives, it’s not the same as law or finance or business where you know that you can go out and become a very successful person. A life in scientific research is unbelievably daunting and challenging, for a lot of reasons, I mean first of all, it’s hard. You go to school for a very long time, but when you get out of school, after 5 years, for doing a PhD, at that point, you’re generally about 30 years old, sometimes older, sometimes younger, you still have to go and do a postdoc, and postdoc salaries are really dictated by the NIH, by the National Institutes of Health. And those salaries are very hard to live on. So a starting postdoc, a 30-year-old person with a PhD might be making $42,000 a year, and you can see why many people maybe wouldn’t choose that path, but even if you do choose that path, and you are gonna live on $42,000 a year in a place like New York City, which—a lot of people do it. Obviously I did it. And at the time, not even making $42,000 a year, I think my salary was $33,000 a year. You know, you still have a very challenging future ahead of you if you’re on the track that where you’re pursuing the traditional scientific career, where you do a postdoc, maybe you do a second postdoc, and then you interview for an assistant professor, tenure track assistant professor job, and get on, you know, the path to a tenured full professor position, maybe even a chairman position. There’s a big drop off at the postdoc stage because making that transition to the next phase is really, really difficult because you have to be unbelievably successful scientifically, you have to publish your work. It would be great if you were funded independently as a researcher, and then those positions for assistant professors, associate professors, full professors, they just don’t come up very often. So it’s hard to be positively reinforced I think, and it’s hard to succeed. I mean it’s hard to compete with the giant pool of postdocs that are out there. Erik Michielsen: Where’s the inflection point, how do you make it easier? Stacie Grossman Bloom: I don’t think it’s going to become easier until the NIH changes their funding structure. The budget for the NIH has been basically flat for a while, which doesn’t give you a lot of incentive to go that route. I mean the success of your grand proposal is not very promising. I think there have to be big changes in the way that we support scientists and fund scientists in this country.

Peter Rowan left his job as a "corporate weenie" in 2010 to live a life with less stuff and fewer expenses and with more time to pursue his dreams. Since downsizing his life, he's begun writing fiction from home (in addition to a teaching gig) so for a bit more privacy, this year he decided to single-handedly convert the family's unused garage into his off-grid writing retreat. Crafting it mostly out of repurposed materials, he decided to heat it using a home-made, recycled can solar panel. After collecting 275 cans (soda, juice, mineral water and beer), he began to drill holes into the ends, glue them together and fit them into a box crafted from plywood and 2 by 4's. Adding a bit of black spray paint and some plexiglass, he created his home-made solar panel. He salvaged a couple of fans from an old computer to create a system for pushing cold air into the solar panel and pushing the hot air out and into his office. In this video, Mary Rowan used her iphone to film her husband's month-long process building a solar heater with recycled cans. Granted Seattle may not be the ideal spot for solar, but Peter says that it seems to heat up the space by about 5 or 6 degrees even on a "crummy day" and when the sun is out it can provide too much heat.
