Computational Imaging: Never Think You've Seen the Last of Anything Lecture by Prof. Peyman Milanfar of University of California Santa Cruz at the TCE conference at Technion-Israel Institute of Technology Fancy cameras used to be the exclusive domain of professional photographers and experimental scientists. Times have changed, but even as recently as a decade ago, consumer cameras were solitary pieces of hardware and glass; disconnected gadgets with little brains, and no software. But now, everyone owns a smartphone with a powerful processor, and every smartphone has a camera. These mobile cameras are simple, costing only a few dollars per unit. And on their own, they are no competition for their more expensive cousins. But coupled with the processing power native to the devices in which they sit, they are so effective that much of the low-end point-and-shoot camera market has already been decimated by mobile photography. Computational imaging is the enabler for this new paradigm in consumer photography. It is the art, science, and engineering of producing a great shot (moving or still) from small form factor, mobile cameras. It does so by changing the rules of image capture — recording information in space, time, and across other degrees of freedom — while relying heavily on post-processing to produce a final result. Ironically, in this respect, mobile imaging devices are now more like scientific instruments than conventional cameras. This has deep implications for the future of consumer photography. In this technological landscape, Google Glass is an example of what is possible and what is still ahead. Glass is the first head-mounted burst-based mobile camera on the market. But this is only a prelude to a much larger story that is just unfolding. Coupled with the ubiquity of devices, open platforms for imaging will inevitably lead to an explosion of technical and economic activity, as was the case with other types of mobile applications. In this wild ride, it is time for clever algorithms, along with dedicated hardware architectures, to take center stage and enable unprecedented imaging capabilities in the user's hands.

The Master´s Degree Programme in Biomedical Imaging aims to train professionals that will have a thorough understanding of diverse imaging technologies along with practical skills in a wide range of imaging methods and applications. Both Finnish and foreign students who have completed a lower university degree equivalent to a Finnish B.Sc. degree are welcome to apply for the programme. The education is given in English. The programme has been assembled on the true imaging strongholds of Turku, and comprises a truly interdisciplinary array of prominent research groups and departments spanning both basic and medical researchers to share advanced imaging techniques in basic cell and molecular biology in disease characterization at the molecular level in patient diagnostics As imaging is a practical science, the programme will include a large practical component, all of which are backed up with solid theoretical background knowledge. Dr. Konofagou received her B.Sc. (Licence) in Chemical Physics from Université de Pierre et Marie Curie, Paris VI in Paris, France and her M.Sc. at Imperial College of Physics, Engineering and Medicine in Biomedical Engineering in 1992 and 1993, respectively. In 1999, Dr. Konofagou received her Ph.D. from the University of Houston in Biomedical Engineering for her work in elastography at the University of Texas Medical School in Houston, TX and then pursued her postdoctoral work in elasticity-based monitoring of focused ultrasound therapy at Brigham and Women's Hospital, an affiliate of Harvard Medical School in Boston, MA. Professor Konofagou is currently an Assistant Professor of Biomedical Engineering and Director of the Ultrasound and Elasticity Imaging Laboratory at Columbia University. She is also a member of the IEEE Ultrasonics, Ferroelectrics and Frequency Control, the Acoustical Society of America and the American Institute of Ultrasound in Medicine. Her main interests are in the development of novel elasticity imaging techniques and applications, and more notably, breast elastography, ligament elastography, myocardial elastography, harmonic motion imaging and focused ultrasound therapy with several close clinical collaborations in the Columbia Presbyterian Medical Center. In the spirit of ideas worth spreading, TEDx is a program of local, self-organized events that bring people together to share a TED-like experience. Fixation refers to a chemical means of killing and preserving cells in a particular physiological state, and in many cases, to preserve morphology. Proper fixation equals preservation of target. * * * * * * * * * * * * * * * The next step of tissue preparation is fixation. (Here, fixation refers to fixed cell systems, such as antibody labeling.) Fixation refers to a chemical means of killing and preserving cells in a particular physiological state, and in many cases, to preserve morphology. The process to achieve this is to crosslink with aldehydes or through solvent fixation. Crosslinking with formaldehyde or glutaraldehyde preserves the structure by crosslinking proteins, that is the amine groups on these proteins, which creates rigidity to the cells or tissue, thus preserving them in that state. However, crosslinking can mask antigen-binding sites. Should an antibody need to bind, antigen retrieval or a protease treatment must take place to unmask or reveal those sites. Additionally, increased autofluorescence, a fluorescence native to the tissue, increases upon crosslinking. Glutaraldehyde has twice the crosslinking power, but it also has twice the autofluorescence. Another possibility is the use of a solvent-based method such as methanol fixation. This method permeabilizes the cells by delipidizing, and opens the cells as well as the antigen-binding site. Some antibodies demand a solvent base, especially if the antibody is a nuclear antigen. However, this method does not the preserve morphology as well as doing the crosslinking with aldehydes. Some membrane-bound components and proteins can be lost. In addition, labeling live cells with a membrane dye such as DiIC18, the components and proteins can with lost with acetone or methanol fixation. Image 8 offers an example of poor fixation. Here, the cells on the left appear fragmented. (Cells that are labeled with a phalloidin to label the F-actin in red.) Cells on the right have been properly fixed. This is evidenced by good morphology and good actin structure. These cells were fixed with formaldehyde. The image also indicates the basic protocol, fixing between 2% and 4%. Methanol-free gives the most success. Formalin, for example, is methanol-stabilized formaldehyde. Most often, the protocol should include the use of methanol-free, EM grade, and fix for 10 to 15 minutes in PBS or another physiological buffer at either room temperature or 37 degrees. Thirty-seven degrees is generally recommend to ensure that, in the last moment before fixation, the cells are still in their proper physiological state. After fixation, wash is recommended. Some cells that are labeled live, technically, are not fixable. Some might be retained. Regarding cells that are labeled live and then fixed, some dyes like nucleic acid stains do not actually crosslink in place, and a fixation will not preserve them. The image on the lower left offers an example of tissue labeled with three colors, three different fluorescent dyes. When put at a warm temperature, 37 degrees for 14 days, the dyes slowly diffuse. This is due to the fact that the dyes are not crosslinked in place, resulting in a washed out appearance. However, some dyes are fixable. The image on the upper right shows two versions of the dye at the top of the gallery. Top left is the dye called FM4-64 on live cells. Top right is the FX, or fixable version of this, on live cells. In the process of fixing the cells, the FM 4-64 (left) is lost, leading to a much-reduced signal. However, the one on the right has amine groups crosslinked in place by aldehydes, and thus retained. It is possible to execute a post-fixation as well, to crosslink, for example, an antibody in place so as not to lose it over time as the slide is archived. The result of proper fixation is preserving the target for later use. Proper fixation equals preservation of target.
