A neat video about how researchers are using a modified HIV virus -- yes, the virus that causes AIDS -- to fight cancer. In the case detailed in the video, a little girl named Emma is now in remission from leukemia as a result of the so-far successful treatment. Emma-HIV-leukemia A reader who's a doctor, Mark Thoma, MD, sent in his summary of the new treatment: If you never thought you'd see the day when you'd appreciate HIV . . . Researchers at the University of Pennsylvania were looking for a way to stop tumor cell growth, or actively kill the tumor cells. They had several patients (adults with chronic lymphocytic leukenia) and kids with acute lymphoblastic leukemia. These patients had failed all previous standard chemotherapy treatments and would all have probably died in less than a 1 month. Researchers at Penn had developed a technique to modify the patients' own T-cells to turn them into what they called "serial killer" cells. These serial killer T-cells could each kill approximately 1000 tumor cells that had the CD-19 antigen on them. This antigen is found on the malignant lymphocytes in these leukemia patients. To produce the serial killers, researchers needed to modify the patients' own T-cells by genetically engineering them to produce an antibody-like protein that would sic them on the tumor cells. To do that, the researchers needed to get at the T-cells' genes. They found that the best way to get this process to work was to use HIV. The HIV naturally seeks out T-cells and binds to them, then injects its own genetic material, functionally taking over the cell. Forcing it to produce millions of copies of HIV before the cell dies. The researchers modified the HIV so that it could no longer cause the infected cells to produce HIV. But it was programmed to cause them to produce the protein that would make them highly specific serial killers. The researchers removed T-cells from each patient, cultured them, added the specially programmed HIV. They let the HIV inject and modify the cells, removed them and infused them into the patients. The results? The modified T-cells were very effective. In one case, the modified T-cells destroyed an estimated two POUNDS of tumor. 12 patients who participated in this small trial (10 adults and 2 children). Nine patients responded and all are still alive, some after almost three years. They show no evidence of any remaining leukemic cells and are generally doing well. The research received additional grant funding and is continuing. The possibility exists that, using modified HIV, other tumor antigens can be successfully targeted and similarly destroyed.

Virus Theory of Cancer There are two kinds of theory: 1. Incomplete theory: Explains what cancer is and how to treat it. 2. Complete theory: Explains what cancer is and how to cure it. Currently the dominating theory is the somatic mutation theory (SMT). Since it does not cure cancer it is incomplete. Rous discovered that sarcoma is caused by a virus, and initiated the viral theory of cancer. At that time neither Rous nor other virologists were interested to cure sarcoma, and their theory remained incomplete. Life evolves within an infinite swarm of viruses. 2.5 million years ago genus Homo confronted bacteria and viruses and had to protect himself. Since viruses plagued life form its very beginning, Homo inherited two protective strategies: 1. Collaboration: Symbiosis. 2. Resistance: Immune system Interaction between cell and virus has two outcomes: 1. Lysis: when virus hijacks cell genome in order to produce virions. Cell dies. 2. Lysogeny when cell hijacks virus genome in order to protect itself from it and from other viruses. Cell lives. The similarity between retrotransposon and retrovirus supports the hypothesis that transposons originated from retrovirus infection. Transposases propel DNA transposons through the genome. Edward Jenner discovered how to treat smallpox by viral interference. SMT is a fatalistic theory. Organism is exposed to disease causing mutations which it cannot resist. This poor theory dragged medicine into its current inconsistency manifested by a toxic targeted therapy. I find it inconceivable that for the last 2.5 million years, organism adapted helplessly to the whims of nature. Which is the essence of neo-Darwinism. Life embraced all these threats creatively, which is the essence of symbiosis. Only recently medicine turned its attention to our symbiosis with gut bacteria (microbiome) . The time has come to embrace virus symbiosis. I believe that autoimmunity diseases are viral infections which might be cured with viral interference.

Viruses are conventionally thought of as the bad guys: antagonists that can cause destruction in our bodies, sicknesses, birth defects, and even kill us. Adam suggests that they may also be beneficial, and possibly required for human survival. Adam is interested in the idea of using viruses as beneficial tools to help treat human genetic diseases such as cancer. He worked three academic years characterizing oncolytic (cancer-killing) poxviruses in the Division of Biology at Kansas State University. He completed a 2015 summer internship in the Department of Entomology at Kansas State University, which resulted in his co-authorship of a scientific manuscript published in the Journal of Experimental Biology. He studied measles virus replication as a 2016 Summer Undergraduate Research Fellow in the Virology and Gene Therapy Track at Mayo Clinic in Rochester, MN. He is a 2016 Barry Goldwater Scholar, which is the United States’ premier honor for college students in mathematics, natural science, and engineering fields. His additional interest in the intersection between science and public policy led to his nomination to represent Kansas State University in the 2017 Harry Truman Scholarship competition. Zika virus destroys brain cancer stem cells - Virus potentially could be used to treat deadly disease. While Zika virus causes devastating damage to the brains of developing fetuses, it one day may be an effective treatment for glioblastoma, a deadly form of brain cancer. New research shows that the virus kills brain cancer stem cells, the kind of cancer cells most resistant to standard treatments. A new study shows that the virus, known for killing cells in the brains of developing fetuses, could be redirected to destroy the kind of brain cancer cells that are most likely to be resistant to treatment. New research from Washington University School of Medicine in St. Louis and the University of California San Diego School of Medicine shows that the virus kills brain cancer stem cells, the kind of cells most resistant to standard treatments. The findings suggest that the lethal power of the virus -- known for infecting and killing cells in the brains of fetuses, causing babies to be born with tiny, misshapen heads -- could be directed at malignant cells in the brain. Doing so potentially could improve people's chances against a brain cancer -- glioblastoma -- that is most often fatal within a year of diagnosis. "We showed that Zika virus can kill the kind of glioblastoma cells that tend to be resistant to current treatments and lead to death," said Michael S. Diamond, MD, PhD, the Herbert S. Gasser Professor of Medicine at Washington University School of Medicine and the study's co-senior author. The findings are published Sept. 5 in The Journal of Experimental Medicine. Each year in the United States, about 12,000 people are diagnosed with glioblastoma, the most common form of brain cancer. Among them is U.S. Sen. John McCain, who announced his diagnosis in July. The standard treatment is aggressive -- surgery, followed by chemotherapy and radiation -- yet most tumors recur within six months. A small population of cells, known as glioblastoma stem cells, often survives the onslaught and continues to divide, producing new tumor cells to replace the ones killed by the cancer drugs. In their neurological origins and near-limitless ability to create new cells, glioblastoma stem cells reminded postdoctoral researcher Zhe Zhu, PhD, of neuroprogenitor cells, which generate cells for the growing brain. Zika virus specifically targets and kills neuroprogenitor cells. In collaboration with co-senior authors Diamond and Milan G. Chheda, MD, of Washington University School of Medicine, and Jeremy N. Rich, MD, of UC San Diego, Zhu tested whether the virus could kill stem cells in glioblastomas removed from patients at diagnosis. They infected tumors with one of two strains of Zika virus. Both strains spread through the tumors, infecting and killing the cancer stem cells while largely avoiding other tumor cells. The findings suggest that Zika infection and chemotherapy-radiation treatment have complementary effects. The standard treatment kills the bulk of the tumor cells but often leaves the stem cells intact to regenerate the tumor. Zika virus attacks the stem cells but bypasses the greater part of the tumor. "We see Zika one day being used in combination with current therapies to eradicate the whole tumor," said Chheda, an assistant professor of medicine and of neurology. To find out whether the virus could help treat cancer in a living animal, the researchers injected either Zika virus or saltwater placebo directly into the brain tumors of 18 and 15 mice, respectively. Tumors were significantly smaller in the Zika-treated mice two weeks after injection, and those mice survived significantly longer than the ones given saltwater. Zika in people would have to be injected into the brain, most likely during surgery to remove the primary tumor. If introduced through another part of the body, the person's immune system would sweep it away before it could reach the brain. The idea of injecting a virus notorious for causing brain damage into people's brains seems alarming, but Zika may be safer for use in adults because its primary targets -- neuroprogenitor cells -- are rare in the adult brain. The fetal brain, on the other hand, is loaded with such cells, which is part of the reason why Zika infection before birth produces widespread and severe brain damage, while natural infection in adulthood causes mild symptoms. Researchers using brain tissue from epilepsy patients showed that the virus does not infect noncancerous brain cells. As an additional safety feature, the researchers introduced two mutations that weakened the virus's ability to combat the cell's defenses against infection, reasoning that the mutated virus still would be able to grow in tumor cells -- which have a poor antiviral defense system -- but would be eliminated quickly in healthy cells with a robust antiviral response. Virus and Cancer In another presentation “Virus Theory of Cancer” I described the relationship between virus and cancer Here I shall explain what do I mean when I say cancer is a viral disease. Tumor is not caused by a virus. It is a byproduct of virus infection. It emerges in a chronically infected tissue. Many viruses are associated with cancer but none displays a function common to all of them. Rous discovered that his cell free filtrate caused a chronic inflammation in which a sarcoma emerged. Only 4% chicken got sarcoma. Clinically RSV causes a chronic muscle inflammation (sarcitis) in which sarcoma emerged. Sarcoma emerged after a long latency, when chicken became weaker. Only in 4% of chicken host resistance became weak enough in order to induce a sarcoma. 96% chicken succumbed to an infection free of sarcoma. I regard Cirrhosis-Hepatoma as a cancer prototype. It illustrates that cancer never appears de novo. It starts as a chronic inflammation of a virus infected liver in which tumor emerges. Cirrhosis is an evolving dysplastic field in which hepatoma emerges. 1. There is nothing in virus genome which is associated with hepatoma. 2. By itself virus does not cause cancer. Hepatom emerges within a chronic inflammation in an immune compromised host (smoldering hepatitis). 3. Patients who recover from hepatitis do not get hepatoma 4. No virus , no cancer. Generally virus has only one goal. To infect a cell and divert its metabolism for its virion production. Which destroys the cell (lysis). Virus has no intention or capability to create a tumor. Tumor is the response of the organism to virus destruction. No virus, no tumor! Viruses are everywhere. In normal and in cancerous tissues. Most of them are our symbionts. Endogenous viruses (HERVs) reside in our DNA and protect us from dangerous exogenous viruses. This phenomenon is called virus interference and may be applied to cure cancer. Think of a virus whenever you spot a tumor within a chronic inflammation. Tumor elimination cannot cure cancer since virus persists. Only virus elimination may cure cancer and this is achieved with virus interference. Although virus does not cause cancer, only virus elimination cures it .

Her aim is to better understand how a common and usually harmless virus called Epstein-Barr virus (or EBV for short), contributes to cancer. Her particular focus is trying to work out how EBV can help cancer cells to cheat death. The overall aim of her work is to design methods to block the interactions between EBV and cellular survival machinery in order to kill cancer cells. Leah is passionate about science communication and public engagement and believes that science should be for everyone and regularly gives talks to researchers and the public about the importance of science and evidence in public life. NTX-010 is in clinical trials for small cell lung cancer and neuroblastoma. CGTG-102 (Ad5/3-D24-GMCSF) is owned by Oncos Therapeutics. GL-ONC1 is owned by Genelux in Phase I clinicla trials. Cavatak is being developed to treat malignant melanoma. Oncorine was approved a few years ago in China for treatment of head and neck cancer - built from H101 virus.
